Lancaster had far better payload, far better range and smaller crew. B-17 had better ceiling, better defensive armament and more rugged construction.
Was the B-17 a good plane?
From such stories, certain airplanes earned a reputation for the ability to bring their crews home during World War II. For instance, there is evidence to suggest that the aircraft often mentioned as the most rugged of World War II—the B-17—was, in fact, stouter than its stablemate, the Consolidated B-24 Liberator.

What is the difference between a B-17 and a Lancaster?
The B-17 had four radial R-1820 engines air cooled. Toward the end of the war 300 Lancasters were produced with Hercules radial engines. The B-17 had better stability and was easier to fly in formation.
What was the best of the three B-17 bombers?
The B-17 is my personal favorite bomber of the war, but again for me the better of the 3 has to be the Lancaster. The He 177 in my opinion was a could have, should have, but did not aircraft. It had great potential but never achieved it. Had she started out with 4 seperate engines, she might have proven to be the better of the 3.
Was the Lancaster bomber better than the Flying Fortress?
The Fort and the Lancaster are very close; the Lancaster could carry a heavier bomb load and was probably every bit as good, or slightly better. But far more B-17s were built and flew, and although I have no stats to back this up, I would guess the Flying Fortress could absorb more punishment and return it’s crews safely.
